Abstract

研究EGb761 (Ginkgo Biloba Extract 761)對周邊神經修復的有效濃度,並利用無血清的培養環境,建立全新周邊神經損傷修復的體外模型,結果將與動物模式作比較。實驗選用Sprague Dawley 老鼠的坐骨神經為史旺氏細胞(Schwann cells)來源,並加入1、10、20、50、100mg/ml 之EGb761。體外培養觀察的結果:(1)當加入EGb761 時,可維持Schwann cell 在無血清環境下的生長,而控制組則明顯較差。(2)經細胞骨架Actin 染色,當EGb761 添加濃度介於1-50μg/ml 之間,細胞形態與控制組無明顯差異,細胞骨架染色結果差異亦不大,100μg/ml可以見細胞之貼附較差,邊緣萎縮,細胞骨架較不明顯。而當劑量增加至250μg/ml時,劑量過高,細胞呈現死亡。實驗並將Schwann細胞植入PLGA神經導管,內填入不同濃度10、 50、100μg/ml 的Egb761,並進行動物植入實驗。
