Abstract

The Geminiviridae family is a large family of plant viruses that has single-stranded DNA genomes and infects a large variety of crop species. In this chapter, we describe a biolistic inoculation protocol that has been successfully used to propagate new species of geminivirus in permissive hosts with total DNA extracted from infected plants. This allows us to directly investigate the biological properties of uncloned and not sap-transmissible geminiviruses. © 2008 Humana Press, a part of Springer Science + Business Media, LLC.
