Abstract

The use of biomimetic polymersomes, poly(2-(methacryloyloxy)ethyl- phosphorylcholine)-co-poly(2-(diisopropylamino)ethyl methacrylate) (PMPC-PDPA) diblock copolymers for efficient DNA encapsulation and delivery was reported. The solvent mixture was evaporated under vacuum, resulting in a copolymer film being deposited on the walls of the vial. The copolymer film was sterilized in an autoclave and then rehydrated under sterile conditions using PBS to form a 0.5% w/w copolymer suspension. Polymersomes encapsulating the green fluorescent proteins (GFP) plasmid were purified through gel permeation chromatography (GPC), using a size exclusion column containing sepharose 4B and using PBS at pH 7.3 to elute the polymersomes. DNA encapsulation was determined by the UV absorption at 260 nm. UV visible spectra confirmed that around 20% of the initial DNA plasmid is encapsulated within the vesicles.
