Glycoprotein non-metastatic melanoma protein B (GPNMB) is a type I transmembrane protein that is also known as Osteoactivin (OA), Dendritic Cell–Heparin Integrin Ligand (DC-HIL) or Hematopoietic Growth Factor Inducible Neurokinin-1 type (HGFIN). GPNMB/OA is expressed in a wide array of normal tissue types including: the bone, hematopoietic system and the skin. Within the bone, GPNMB/OA has been shown to promote the differentiation of both osteoclasts and osteoblasts. GPNMB/OA is also readily detectable in immune cells, such as macrophages and dendritic cells, and has been shown to functionally impair T-cell activation. Within the skin, GPNMB/OA has been proposed to be expressed specifically in melanocytes, while others suggest a broader pattern of expression that includes keratinocytes, melanocytes and Langerhans cells.
