APLICAÇÃO DE ANTIGÉNIOS RECOMBINANTES NO DIAGNÓSTICO SEROLÓGICO DA TOXOPLASMOSE

Abstract

Toxoplasma gondii é um protozoário oportunista pertencente ao filo Apicomplexa, que pode desencadear graves complicações em pacientes imunocomprometidos e gestantes. O diagnóstico da toxoplasmose é geralmente feito através da observação do parasita em amostras biológicas ou pela deteção de antigénios IgM e IgG anti-T. gondii específicos no soro dos doentes. A melhoria das técnicas de diagnóstico da toxoplasmose e a distinção entre as fases da infeção pode ser conseguida através da utilização de proteínas recombinantes. Este estudo tem como objetivo a utilização da proteina recombinante do micronema MIC3 (que desencadeia uma resposta imunitária específica no hospedeiro) no serodiagnóstico da toxoplasmose. O RNA total de Toxoplasma gondii foi isolado utilizando o método de Tri-reagent e os genes que codificam a proteina MIC3 foram amplificados. A proteina recombinante foi clonada no vetor pNZY 28 por TA cloning e no vector de expressão pLATE 31, através da tecnologia de clonagem independente de ligase (LIC). As amostras foram submetidas a sequenciação e demonstraram homologia com a sequência da proteina MIC3 de T. gondii armazenada na base de dados do GenBank. Um estudo de expressão com IPTG foi executado em diferentes estirpes de E. coli BL21 (DE3): Star, XJB, RIPL e PlysS transformadas com o vetor pLATE 31. Uma vez que este vetor permitiu a produção do antigénio recombinante MIC3 com uma cauda de polihistidina, MIC3 foi purificado por cromatografia de alta-afinidade com iões de níquel imobilizados, testando vários tampões com diferentes valores de pH. As amostras eluídas foram analisadas por ELISA, eletroforese SDS-PAGE e por leitura das absorvâncias a 280 nm, de forma a verificar qual a estirpe mais produtora de proteina recombinante e o melhor método de purificação do antigénio recombinante MIC3. Também foram utilizadas técnicas de dot-blot e western-blot para demonstrar a presença da proteina recombinante MIC3 e os resultados mostram que E. coli BL21 XJB (DE3) e o tampão Fosfato de Sódio pH 7.5 utilizado na purificação são a melhor combinação para purificar a proteina recombinante. ; Toxoplasma gondii is an opportunistic apicomplexan protozoon that can cause devastating disease in immunosuppressed patients and congenital infection. The diagnosis of toxoplasmosis is usually done by observing the parasite in biological samples or by the detection of specific IgM and IgG against T. gondii antigens in the patient’s serum. The improvement of toxoplasmosis diagnostic techniques and the differentiation between the infection stages can be achieved using recombinant antigens. This study aims to use the micronemal protein MIC3 (elicits a strong specific host immune response) recombinant antigens in the serodiagnosis of toxoplasmosis. Total Toxoplasma RNA was isolated using the Tri-Reagent method and genes encoding MIC3 were amplified. The recombinant protein was cloned into the cloning vector pLATE 28 by TA cloning, and into the expression vector pLATE 31, through ligation independent cloning (LIC) technology. The samples were sequenced and showed homology with the sequence of T. gondii MIC3 protein, stored in GenBank’s database. An expression study with IPTG was performed in different E. coli BL21 (DE3) strains: Star, XJB, RIPL and PlysS transformed with pLATE 31. Since this vector enabled the production of the recombinant antigen MIC3 with a polyhistidine tail end, MIC3 was purified by high-affinity chromatography with immobilized nickel ions, testing several tampons with different pH values. The eluted samples were analysed by ELISA, SDS-PAGE electrophoresis, by reading it’s absorbance at 280 nm, in order to show which strain would produce more recombinant antigens and which method would be the best to purify the recombinant antigen MIC3. A dot-blot and a western-blot were also performed to show that the recombinant protein MIC3 was present and results show that E. coli BL21 XJB (DE3) and the tampon Sodium Phosphate pH 7.5 used in the purification is the best combination to purify the recombinant protein and that the recombinant.
